1. Summary
Aflatoxins are toxic metabolites of a class of fungi such as Aspergillus flavus and Aspergillus parasiticus. They are highly carcinogenic and are found in peanuts, grains, nuts, cottonseed, and other food, animal feed and other related products. The naturally contaminated aflatoxin is mainly aflatoxin B 1 . In 1993 , aflatoxin was classified as a Class I carcinogen by the World Health Organization ( WHO ) Cancer Research Institute , and it is a highly toxic and highly toxic substance. It has been shown to be very harmful to tissues and organs such as the liver and kidneys of humans and animals. Aflatoxin-producing molds (such as Aspergillus flavus, Aspergillus parasiticus, etc.) are spread all over the world. Aflatoxin contamination can occur in the process of plant growth, harvesting, processing, storage, transportation, etc. It is best to detect the source of pollution in time. A method of aflatoxin contamination. China requires that aflatoxin B 1 in peanuts and their products should not exceed 20 μg / kg. International CAC stipulates that the total aflatoxin (AFT) in peanuts and their products should not exceed 15μg / kg . The total amount of aflatoxin in edible grains in the EU should not exceed 4μg / kg , and aflatoxin B 1 should not exceed 2 μg / kg . The iCheck® Aflatoxin Quantitative Card can quickly and quantitatively detect the total amount of aflatoxin from 0-25 ppb in peanuts .

3. Equipment required for testing
1 ) 200μL , 1000μL micropipettes ( 1 each)
2 ) iCheck ® quantitative detector ( with zero calibration sheet )
3) Multi-purpose rotary shaker
4) Timer 5) Electronic scale 6) pure water, or double distilled water
7) Extract: methanol + water ( 70+30 )
8) Funnel 9) Qualitative filter paper 10) 50 mL centrifuge tube
4. Preparation before testing and calibration of the instrument
1) Return the quick check card, buffer and sample to be tested to room temperature (25 °C ± 2 °C) for 30 minutes before use.
2 ) Equipment calibration (recommended once a month): Insert the calibration film into the quantitative quick test chamber and correct it in “Settings”.
3 ) Equipment calibration: put the AFT QR Code of the same batch number into the quantitative quick checker card slot and read in the QR code.
4) According to the number of samples to be measured, open the package, take out the quick check card and AFT micropores, and place the AFT micropores on the microporous rack.
5. Sample processing and testing
1) Weigh 5.0 g of crushed (20 mesh sieved) peanut samples in a 50 mL centrifuge tube, add 25.0 mL of 70% methanol water extract, and place on a rotary shaker for 5 minutes ( or use a high-speed homogenizer). For 1 minute, or vigorously shake by hand for 3 minutes ) , filter with qualitative filter paper.
2 ) 120 μL of the dilution buffer CSB-4 was placed in the AFT microwell, 30 μL of the filtrate was added , and the mixture was pipetted 5 times, and 100 μL was added to the sample hole of the quick test card.
3 ) After reacting at 37 °C ( ±2 °C ) for 10 minutes, immediately put the quick check card into the quantitative quick tester and click the “ analysis ” reading, which is the actual concentration of the sample.
6. Result determination
1 ) Detection range: 0-25ppb, the limit of quantification is 3.0ppb.
2 ) This test card is a quantitative quick check card. If there is no red line on the control line ( C ), it indicates that the quick check card is invalid.
3 ) The total aflatoxin determination value is greater than 80% of the national or internal limit value (for example, the total aflatoxin value of peanut is greater than 16ppb ), it is recommended to re-measure it once. If it is still greater than these values, please use the instrument method for confirmation analysis.
4 ) If the measured value is 25 ppb , dilute the filtrate of the sample extract with 70% methanol water, and finally quantify the reading in the rapid tester by the dilution factor, which is the actual concentration of the sample.
7. Precautions
1) The quick check card is used once during the warranty period. Store the refrigerator at 2-8 °C until it is equilibrated to room temperature (25 °C ± 2 °C) before use.
2) Beware of the moisture in the quick check card, the aluminum foil packaging bag is damaged or the quick check card is damp, the test card should not be used.
3) Try not to touch the white film in the center of the quick check card.
4) Avoid direct sunlight during all incubations.
5) The small bag in the aluminum foil packaging bag is a desiccant and should not be taken into account.
6) Exceeding the shelf life cannot be used.
