Behind the cloned monkey: Dr. Tu's refining cell "nuclear" first person

Release date: 2018-06-11

On January 25th, the cover article published online by Cell magazine sensationalized the world: On November 27, 2017, the world's first individual cell cloned monkey "中中", which was cultivated by the team of the Institute of Neuroscience of the Chinese Academy of Sciences, was born, the same year 12 On the 5th of the month, the second individual cell cloned monkey "Huahua" was born. This achievement marks the first time in China to open a new era of somatic cloned monkeys as experimental animal models.

At the results conference held in Shanghai on January 25th, Sun Qiang, the author of the article and a researcher at the Institute of Neuroscience of the Chinese Academy of Sciences, said that the somatic cells of macaques were first cultured in vitro, the nuclei were removed, and the other one that had removed the nuclei was injected. In the oocytes of macaques, this cloned embryo is transplanted into the monkey monkey palace. The macaques produced are the somatic cloned monkeys "Zhongzhong" and "Huahua".

It is easier said than done, but it is a problem that scientists all over the world have been unable to overcome in the past 20 years. Just a “go to nuclear” step, the first author of the article, Liu Zhen, a postdoctoral fellow at the Institute of Neuroscience of the Chinese Academy of Sciences, practiced for many years.

Through the microscope, the polarized light flashes and flashes, causing pain in the eyes. Since the nucleus is not easily identifiable, the nucleus can be exposed under the microscope only under polarized light. Liu Zhen holds only 10 micron thick glass needles, carefully passes through the macaque egg cells with a diameter of only 100 micrometers, finds the nucleus of sesame size in the line of sight, gently removes it, and the whole process is completed in one go. In order to minimize cell damage and increase embryo survival, the overall operating time must be as short as possible.

For rhesus somatic cell cloning, cell "nucleation" is a very difficult technique. Moreover, compared with mouse oocytes, macaque oocytes are much smaller and more difficult to operate. From the second year of the master's degree, the instructor Sun Qiang handed over the technical responsibility of “going to the nuclear” to Liu Zhen. After many years of hard work, Liu Zhen can complete the “nucleation” process of 6 macaque oocytes in 52 seconds. Similarly, Liu Zhen, who was in the process of “injecting the nuclear”, was also trained to complete the operation in just 15 seconds.

In the eyes of Sun Qiang, Liu Zhen is a well-deserved "world champion" in the technology of "nuclear" and "nuclear" cells. Liu Zhen is a native scientist who was trained by the Chinese Academy of Sciences and has no overseas study experience.

In 2010, Liu Zhen came to the Institute of Neuroscience of the Chinese Academy of Sciences to study for a master's degree. Two years later, he followed the mentor to develop a somatic cell clone monkey, a world-class difficulty project. Due to the strong research foundation of the Institute's primates, and Liu Zhen's special affairs, Liu Zhen, who had been able to go to the top research institutes in the United States, stayed in the scientific research until now. However, in the view of Academician Pu Muming, director of the Institute of Neuroscience of the Chinese Academy of Sciences and the Center for Excellence in Brain Science and Intelligent Technology, it is hard to imagine young scientists studying abroad. "They generally don't choose a highly risky project, but choose 'short and fast', send one or two high-quality articles, and graduate smoothly and return home."

Academician Pu Muming said at the press conference that Chinese scientific research needs to be a leader in the world science from "gradual" innovation to a major breakthrough, and the cultivation of local youth innovative talents is of paramount importance. However, what deplores him is that the current domestic policy orientation is not conducive to this. Colleges and research institutes are more inclined to recruit young scientific and technological talents with overseas study experience, and the funding for young scientists who are studying abroad is even greater. This phenomenon has not improved significantly over the years. In fact, the young scientific talents cultivated locally are not worse than those returned from overseas. Liu Zhen is such an example.
