Rapid and accurate determination of ethanol concentration in blood: headspace-gas chromatography

Headspace-gas chromatograph analysis of the determination of ethanol in blood, the method has been improved for many times, has been widely used in countries around the world, mainly used to test the concentration of ethanol in the blood of drunk drivers. Determination of Ethanol in Blood by Headspace-Gas Chromatography

1, sample preparation

Ethanol standard stock solution: 10g / L aqueous solution, add Ili pour mercury iodide crystals, and store in cold storage.

Internal standard solution: φ = 1.0% aqueous solution of n-propanol as a stock solution, diluted to 0.25% with water .

Standard solution: Dilute the ethanol stock solution by 10 times and then take 1 ml of the solution into a 10 mL headspace vial, and add 1 mL of the internal standard solution to seal quickly. This sample was used to determine the correction factor.

Blood sample: 1 mL of well-mixed blood sample was transferred to a 10 mL headspace vial, and 1 mL of internal standard solution was added to seal quickly.

2, analysis conditions

Column: 2m × 2mm glass packed column with 5% PEG-20M/CarbopackB ( 60~80 ) mesh.

Column temperature: 75 °C (the column temperature is suitably lower when using a capillary column).

Packed column inlet, 150 °C; FID detector, 200 °C.

Carrier gas: helium or nitrogen, 30mL/min .

3, headspace conditions

The equilibrium temperature is 50 °C; the equilibrium time is 10 min ; the valve, the delivery tube and the quantitative tube temperature are 80 °C; the pressure is 13.8 kPa ; the pressurization time is 0.15 min ; the filling time is 0.15 min ; the pressure equilibration time is 0.15 min ; and the quantitative tube is 1.0 mL .
